Abstract: Recent advances in serial ultrathin-section techniques for electron microscopy (EM) allow large-scale dense neurite reconstruction at the synapse level. By means of serial block-face EM and connectomic analysis, we have uncovered higher-order circuit structures in the sensory cortex layer IV as well as novel connections in the auditory nervous system. Besides, I will review recent methodological developments in quantitatively mapping synaptic plasticity and innervation specificity in 3D EM datasets.
